Less toxic treatment strategies for testicular germ cell tumor (TGCT) patients are needed, as overtreatment is a concern due to the long-term side effects of platin-based chemotherapy. Although clinical benefit from classical hypomethylating agents has to date been limited, TGCTs show an abnormal DNA methylome indicating the potential of treating TGCTs with hypomethylating drugs. We tested, for the first time in TGCT cell lines, a new synthetic flavonoid compound (MLo1302) from the 3-nitroflavanone family of DNA methyltransferase (DNMT) inhibitors. We show that MLo1302 reduces cell viability (including of cisplatin resistant cell line NCCIT-R), with IC50s (inhibitory concentration 50) within the nanomolar range for NCCIT and NTERA-2 cells, and proved its cytotoxic effect. Exposure to MLo1302 reduced DNMT protein expression, similar to decitabine, and showed a partial effect in cell differentiation, reducing protein expression of pluripotency markers. RT2 profiler expression array indicated several dysregulated targets, related to activation of apoptosis, differentiation, and cell cycle arrest. We validated these data by showing increased apoptosis, increased protein expression of cleaved caspase 8 and activated caspase 2, and reduced proliferation (BrdU assay), with increase in CDKN1A and decrease in MIB-1 expression. In order to evaluate the latter in the context of process model comprehension, this paper presents the results from ongoing research, in which gaze data from experts are used as Eye Movement Modeling Examples (EMMEs) to convey visual observation capabilities to novices. Compared to prior results, the application of EMMEs improves process model comprehension significantly for novices. Novices achieved in some cases similar performances in process model comprehension to experts. The study's insights highlight the positive effect of EMMEs on fostering the comprehension of process models.The hermeticity performance of the cavity structure has an impact on the long-term stability of absolute pressure sensors for high temperature applications. In this paper, a bare silicon carbide (SiC) wafer was bonded to a patterned SiC substrate with shallow grooves based on a room temperature direct bonding process to achieve a sealed cavity structure. Then the hermeticity analysis on the SiC cavity structure was performed. The microstructure observation demonstrates that the SiC wafers are tightly bonded and the cavities remain intact. Moreover, the tensile testing indicates that the tensile strength of bonding interface is ~8.01 MPa. Moreover, the quantitative analysis on the airtightness of cavity structure through leakage detection shows a helium leak rate of ~1.3 × 10-10 Pa?m3/s, which satisfies the requirement of the specification in the MIL-STD-883H. The cavity structure can also avoid an undesirable deep etching process and the problem caused by the mismatch of thermal expansion coefficients, which can be potentially further developed into an all-SiC piezoresistive pressure sensor employable for high temperature applications.In this study, tap water is magnetized by a self-developed device. The conductivity and evaporation of magnetized water (MW) at different temperatures are tested to demonstrate the magnetization mechanism. The results show that the conductivity and evaporation of the magnetized water increase to different degrees compared with regular tap water (RW). The maximum increase in conductivity is 10.6%, and the maximum increase in evaporation is 25.6% and 16.7% at 50 and 80 °C, respectively. Cement grout samples with water-cement ratios (w/c) of 0.5 and 1.0 were prepared with magnetized water. The stability of the cement grout mixed with RW and MW under different magnetic conditions is tested. The compressive strength and SEM images of the hardened cement grout samples mixed with RW and MW (under optimum magnetic conditions) are compared. The cement grout prepared with MW has a higher stability compared to that of the control specimen, and the relative change of bleeding volume of the MW cement grout with w/c = 0.5 and 1 is 67.69% and 24.36%, respectively. MW has a positive effect on the consolidation compressive strength of cement grout. SEM images show that hardened cement grout with MW has more hydrate crystals, more compact filling space between cement particles, more contact points, and fewer voids compared to that of RW. The influence mechanism of MW on the stability of cement grout is analyzed, which provides a theoretical basis for the application of MW in the field of grouting engineering.
